Spectrophotometers

A spectrophotometer is an analytical instrument used to measure the intensity of light at different wavelengths across the electromagnetic spectrum. It operates based on the principle that different substances absorb, transmit, or reflect light to varying degrees depending on their molecular structure. This makes spectrophotometers invaluable in fields like chemistry, biology, physics, and environmental science for analysing and quantifying compounds.

The device typically consists of a light source, a monochromator (which selects specific wavelengths of light), a sample holder, a detector, and a readout system. The light source emits light across a wide range of wavelengths, and the monochromator narrows it down to a specific wavelength of interest. This light is directed onto the sample, and the detector measures how much light passes through the sample or is absorbed by it. The data obtained from the detector is then converted into an absorbance or transmittance spectrum, which is used to identify the chemical composition of the sample.

Spectrophotometers come in various types, such as UV-Vis spectrophotometers (which measure light in the ultraviolet and visible range), IR spectrophotometers (for infrared light), and fluorescence spectrophotometers (which measure emitted light after excitation). UV-Vis spectrophotometers, for instance, are commonly used for determining the concentration of substances like proteins, nucleic acids, and organic compounds in solution.

One key application is quantitative analysis, where the absorbance of a sample at a specific wavelength correlates with its concentration, following Beer-Lambert Law. This allows spectrophotometers to be used in clinical diagnostics, pharmaceutical quality control, environmental monitoring, and research. Overall, spectrophotometers are essential tools for both qualitative and quantitative analysis, providing highly accurate and reproducible results.
